چکیده
Objective: Lung cancer is a refractory tumor with poor five-year survival rate. Ebracteolatain A (EA), key acetyl-phloroglucinol derived from the traditional Chinese herb Langdu, has been reported to have anti-tumor activity in human breast cancer. However, therapeutic efficacy and underlying mechanism of lung remain be elucidated. In present study, we intended evaluate whether EA exhibits anti-lung elucidate mechanisms. Methods: ATPlite clonogenic assays were used assess inhibiting effect on two cell lines vitro. FACS was conducted determine cell-cycle profiling cells upon treatment EA. Western blot performed detect level related proteins EA-treated cells. RNA silencing other experiments identify molecular EA-triggered arrest. Furthermore, xenograft model constructed potential vivo mice. Results: significantly inhibited growth both vitro vivo. Mechanistically, EA-induced G 1 phase arrest mediated by accumulation p21 p27. Conclusion: Our findings revealed mechanisms, providing solid scientific basis for as an attractive choice therapy.

SIRT2 inhibits non-small cell lung cancer cell growth through impairing Skp2-mediated p27 degradation
Skp2 is a component of the E3 ubiquitin ligase which promotes the ubiquitination-associated degradation of a cyclin-dependent kinase inhibitor, p27, resulting in increases in non-small cell lung cancer (NSCLC) cell growth. We recently showed that down-regulation of Sirtuin deacetylases 2 (SIRT2) in NSCLC increased cancer cell growth through suppressing p27. However, the underlying mechanisms re...
متن کاملGatifloxacin Induces S and G2-Phase Cell Cycle Arrest in Pancreatic Cancer Cells via p21/p27/p53
Pancreatic cancer, despite being the most dreadful among gastrointestinal cancers, is poorly diagnosed, and further, the situation has been aggravated owing to acquired drug resistance against the single known drug therapy. While previous studies have highlighted the growth inhibitory effects of older generation fluoroquinolones, the current study aims to evaluate the growth inhibitory effects ...
متن کاملAnti-cancer effect of novel PAK1 inhibitor via induction of PUMA-mediated cell death and p21-mediated cell cycle arrest
Hyper-activation of PAK1 (p21-activated kinase 1) is frequently observed in human cancer and speculated as a target of novel anti-tumor drug. In previous, we also showed that PAK1 is highly activated in the Smad4-deficient condition and suppresses PUMA (p53 upregulated modulator of apoptosis) through direct binding and phosphorylation. On the basis of this result, we have tried to find novel PA...
متن کاملPolydatin inhibits growth of lung cancer cells by inducing apoptosis and causing cell cycle arrest
Polydatin (PD), a small natural compound from Polygonum cuspidatum, has a number of biological functions. However, the anticancer activity of PD has been poorly investigated. In the present study, thiazolyl blue tetrazolium bromide assay was used to evaluate the inhibitory effect of PD on cell growth. Cell cycle distribution and apoptosis were investigated by flow cytometry. In addition, the ex...
